Protocol 5 – DON doses 2 and 3

For the second dose of this protocol (Friday, June 28th), I stuck with the enteric-coated capsules. This is becoming my new standard unless I switch to subcutaneous injections, which seems unlikely, at least not before my following scan. If my breast tumour disappeared back in the fall of 2023 when I was taking DON orally (and losing half to stomach acids), then bypassing the stomach should theoretically work even better. I believe the issue at this point isn’t the absorption of DON but rather my challenge in keeping my glucose levels as low as I’d like.

I did the same for dose two as for dose one. I took 0.6mg/kg. I included my morning meds, which consisted of 500mg of Metformin, but I took them much earlier so as not to impact my gastric emptying speed. I took those at 07:30, and then, at 11:00, I took the DON. I was in the hyperbaric oxygen tank by noon and experienced no side effects, which confirmed my suspicion that my nausea during the first dose was due to the capsule opening in my stomach.

On July 1st for my third DON day, I increased to 1mg/kg. This is close to my former standard dose from previous protocols, but now I am bypassing the stomach. In theory, I’m getting more into my bloodstream as if taking twice as much. I had no side effects! Ok, so my epithelial cells are rock stars, I guess?!

At this point, I need to evaluate the benefit of increasing the dose further. I don’t think it’s necessary since I succeeded at 1.1mg/kg taking it orally without a fancy capsule. However, I have this nagging fear (without any objective evidence) that it won’t be as effective if I don’t take enough. Perhaps taking more would result in more cancer cell death, but I think I might just be overthinking it.

My main concern should be managing my glucose levels, as that’s proving incredibly difficult. I’m trying to keep them between 3.2-3.6 mmol, which has been nearly impossible without fasting. My next step is to plan five daily snacks with a 4:1 ratio and add dapagliflozin. For my next dose this Friday, I plan to stay close to 1mg/kg, as there might be some cumulative damage happening, and at some point, I might experience significant side effects.

Pretty dull when there is no vomiting, isn’t it?!
